Depending on how far it is open...it affects it some...but it's probably negligible since the sunroof has a little spoiler that pops up which facilitates diminishing wind buffering as well as helping to direct air over the top of the car and prevents it from creating a low pressure area inside the cabin (unlike what happens in the bed of a pick-up truck).

My commute is Portland to Seattle, so about 3 hours with about 2 hours at 70+mph. Absolutely no difference in mileage with sunroof open and sunroof closed. Now noise can get to be annoying after a couple of hours of having the sunroof open and all the traffic.
